The ground was scheduled to be ready for the start of the 2018/19 Premier League season, however Tottenham have requested to play their first scheduled home game against newly-promoted Fulham at Wembley on August 18.
Mauricio Pochettino's side played at the national stadium for the entirety of last season while their new home underwent a major overhaul.
Harry Kane, Dele Alli et al will now walk out at the ground for the first time when they face Liverpool on September 15.
Manager Pochettino said: “We are so excited to have the date of the first Premier League match at our new stadium.
“We discussed the timings and the ability to play away games and I think it is a good solution to have an earlier home game.
The club is aiming to open the stadium with test events, expected to include an open training session for the first team, taking place in advance of the Liverpool fixture.
