明 細 書
高分子電解質錯体抗菌剤及び抗菌性材料 技術分野
本発明は、 高分子電解質錯体からなる抗菌剤及びその高分子 電解質錯体を担持してなる抗菌性材料に閧する。 背景技術
正電荷を有する有機化合物は、 それが低分子化合物であるか 高分子化合物であるかを問わず、 抗菌性を示すことが知られて いる。 この性質を利用した応用技術は医療分野から一般衣料品 にまで広く普及している。 例えば、 塩化ベンザルコニゥム等に 代表される第 4級アンモニゥム化合物は水溶性であるため、 そ れ自体が殺菌 ·消毒用液として用いられている。 しかし、逆に、 水溶性であるために抗菌剤としての使用範囲が限られている。
また、 合成高分子成形品の表面にァニオン性基を導入し、 続 いて第 4級アンモニゥム塩基で処理することにより、 長期間持 続的に抗菌性を示す材料を得て、 エアーフィルターや透析用の 沪過材として用いることも行なわれている。 更に、 この技術を 繊維材料に応用し、 抗菌性を有する衣料品あるいは創傷保護剤 として利用されている。
例えば、 高分子材料表面に第 4級アンモニゥムを導入し、 抗 菌性を持続的に示す素材を得る手段として、 特開昭 5 4— 8 6 5 8 4号公報には、 酸性基含有モノマーを重合させて得た高分 子物質を第 4級アンモニゥム塩基の水溶液と接触させる方法が 記載されている。 また、 特開昭 5 9— 1 6 4 3 4 2号公報には、 ァニオン性基含有ビニルモノマーをグラフト重合等によって合 成高分子成形品の表面に導入し、 続いて第 4級アンモニゥム塩 基で処理する方法が開示されている。
しかしながら、 従来法では、 抗菌性を発現する第 4級アンモ ニゥム塩基の導入率が必ずしも十分なものではなく、 持続性も 満足できるものではなかった。 更に、 その製造方法も煩雜であ つた 0
本発明者は、 正荷電有機化合物の抗菌性を維持したままで溶 媒(特に水)不溶性の性質を有し、 しかも持続的な抗菌作用を 有する材料を得ることを目的に鋭意研究を行なったところ、 特 定のァ二オンボリマーと特定のカチオンボリマーとを反応させ て得られる高分子電解質錯体によってこれらの目的を達成する ことができることを見出した。 本発明はかかる知見に基づくも のである。 発明の開示
従って、 本発明は、 (A )繰返し単位中に N+原子を含有す るカチオンポリマーと (B )繰返し単位中に一 C O O一基、 - S 03一基又は一 P 03一基を含有するァニオンポリマーとを反 応させることによって得られる高分子電解質錯体を含有するこ とを特徴とする抗菌剤に鬨する。
また、 本発明は前記の高分子電解質錯体を担体上に担持する ことを特徴とする抗菌性材料にも鬨する。

本発明で用いることのできる高分子電解質錯体
( p o l y e 1 e c t r o l y t e c omp l e x :以下、 PECと称することがある) は、 それ自体公知の物質である。 高分子電解質錯体 ( PEC ) は、 例えば特開昭 49— 8581 号公報に記載されているように、 正荷電を有する高分子電解質 であるカチオンポリマーの溶液と負荷電を有する高分子電解質 であるァニオンポリマーの溶液とを混合することにより瞬時に 形成することができる。 こうして得られた P E Cは、 特殊な 3 元系溶媒(例えば、 特定の組成からなる、 水 Zアセトン Z低分 子塩) には溶解するが、 一般的な溶媒には不溶性である。 P E C膜は、種々の低分子化合物に対して高い透過性を示すので、 透析膜として用いることができる。 P E Cは、 出発ポリマー (高分子電解質) の種類、 それらの混合比、 調製条件などによ り、 多様な性質を有する各種の材料を提供することができるが、 P E Cが抗菌性を有することは従来知られていなかった。
本明細書において、 炭素数 1〜3個のアルキル基は、 例えば、 メチル基、 ェチル基、 又は n—若しくは i一プロピル基である。 炭素数 1〜 4個のアルキル基は、 前記炭素数 1〜3個のアルキ ル基の他に、 例えば、 n—、 i―、 s—、 又は t—ブチル基で ある。 炭素数 1又は 2個のアルキレン基は、 例えば、 メチレン 基、 エチレン基又はェチリデン基である。 炭素数 1〜1 0個の 直鎮又は分枝鎖のアルキレン基は、 例えば、 メチレン基、 ェチ レン基、 プロピレン基、 トリメチレン基、 テトラメチレン基、 ペンタメチレン基、 へキサメチレン基、 ヘプタメチレン基、 ォ クタメチレン基、 ノナメチレン基、 デカメチレン基: ェチルェ チレン基、 又はェチルトリメチレン基である。 炭素数ら〜 1 8 個の直鎖若しくは分枝鎖のアルキル基は、 例えば、 炭素数 6〜 1 8個の直鎖アルキル基、 炭素数 1〜3個の直鎖若しくは分枝 鎖のアルキル基 1個又はそれ以上で置換された炭素数 6〜 1 5 個のアルキル基、 特には、 炭素数 1〜3個の直鎖アルキル基 1 個で置換された炭素数 6〜1 0個のアルキル基である。 ァリー レン基は、 例えば、 フエ二レン基又はナフチレン基である。 力 ルボン酸基又はスルホン酸基の塩は、 例えば、 アルカリ金属 (例えば、 ナトリウム、 カリウム〉 、 又はアルカリ土類金属 (例えば、 カルシウム、 マグネシウム〉 との塩である。 また、 スルホン酸基若しくはその塩を含有するァリール基は、 例えば、 スルホフェニル基である。
出発材料のカチオンボリマー (A〉 及びァニオンポリマー ( B ) 中に存在する対イオンは、 両者の反応の結果、 P E C生 成物中には存在しなくなる。 従って、 両者の反応を阻害しない ものならいかなるイオンであってもよい。 好ましい対イオンは ハロゲン原子イオン、 特には塩素イオン、 臭素イオン又はヨウ 素イオンである。
カチオンポリマ一 (A ) とァニオンボリマー ( B ) との反応 によって得られる P E Cは、 カチオンポリマーの N+部位とァ 二オンポリマーの酸 (例えば、 カルボン酸、 スルホン酸、 リン 酸) 部位とがクーロン力で次々に結合した楕造を有する。 すな わち、 高分子同士がイオン結合により橋架けされ、 溶媒不溶性 のゲルとなっている。
例えば、 一般式 ( I ) で表されるカチオンポリマー ( a 1 ) と一般式 (IV ) で表されるァニオンポリマー ( b 1 ) とから生 成される P E Cは以下の一般式 (VI ) で表される。

本発明で抗菌剤として用いることのできる P ECは、 溶媒不 溶性の固体であればよく、 その平均分子量は特に制限されるも のではないが、 一般には、 イオン席の数にして、 10〜
1000、 好ましくは、 20〜100である。 出発材料である カチオンポリマー ( a 1 )〜 ( a4 ) 、 並びにカチオンボリマ 一 ( b 1 )〜 ( b 3 ) の平均分子量も特に制限されるものでは ないが、 好ましい平均分子量範囲は、 一般式 ( I ) で表される カチオンボリマー ( a 1〉 では、 mが 5〜500 (特には 5〜 100 ) であり、 一般式 (II) で表されるカチオンボリマー ( a 2 ) では、 nが 10〜 1000 (特には 10〜500 ) で あり、 一般式 (III) で表されるカチオンポリマー ( a3 ) では、 rが 10〜; L 000 (特には 10〜500 ) である。 これに対 し、 一般式(IV)で表されるカチオンポリマー (b 1 )では、 qが 10〜: L 000 ( には 10〜500 ) であり、 一般式 (V) で表されるカチオンポリマー ( b 2 ) では、 Pが 10〜 1000 (特には 10〜500 )である。
前記一般式( I ) のカチオンポリマー ( a 1 ) の具体例を挙 げれば、 第 4級ポリエチレンィミンク口ライド、 ポリ (N, N, Ν' , N' —テトラメチルーアルキレン一 Ρ—キシレンジアン モニゥムジクロライド) 、 ポリ (Ν, Ν, N' , N' —テトラ メチル一アルキレンージアンモニゥムジクロライド) 、 ポリ (Ν, Ν—ジメチルー 3—ヒドロキシプロピルアンモニゥムク 口ライド) 、 ポリ ( 2—ヒドロキシー 3—メタクロィルォキシ プロピルトリメチルアンモニゥムクロライド) 、 ポリ ( 2—メ タクロイルォキシェチルトリメチルアンモニゥムクロライド) 、 ポリ (グリシジルトリメチルアンモニゥムクロライド) 、 ポリ [ (ジメチルイミニォ)エチレン(ジメチルイミニォ) メチレ ン一 1 , 4一フエ二レンメチレンジクロライド] [一般に、 2 Xと称される] 、 ポリ [ (ジメチルイミニォ)へキサメチレン (ジメチルイミニォ) メチレン一 1 , 4—フエ二レンヌチレン ジクロライド] [一般に、 6Χと称される] 、 ポリ [ (ジメチ ルイミニォ)へキサメチレンクロライド] [一般に、 6, 6と 称される] 、 ポリ (Ν—ェチル—4一ビニルピリジニゥムブロ マイド) 、 ポリ (ジメチルジァリルアンモニゥムクロライド) 等である。
前記一般式(Π) のカチオンポリマー ( a2 ) の具体例を挙 げれば、 ポリ (ビニルベンジルトリメチルアンモニゥムクロラ イ ド) 、 ポリビニルピリジニゥムクロライ ド、 ボリ (N—ベン ジル一 4一ビニルピリジニゥムクロライ ド) 等である。
前記一般式 (III) のカチオンポリマー ( a3 ) の具体例を挙 げれば、 ボリリジン、 ボリアルギニン又はこれらポリマーを構 成する単量体のコポリマー、 更に、 これら単量体とグリシン、 ァラニン、 フエ二ルァラニン、 チロシン、 ノ S'リン、 ロイシン、 イソロイシン、 セリン、 トレオニン、 メチォニン、 システィン、 ヒスチジン、 プロリン、 及び/又はトリアトファンなどとのコ ポリマーである。
カチオン性多糖類 ( a4 ) の具体例を挙げれば以下の通りで ある。
( 1 ) キトサン及びその誘導体:
式中、 R66は水素原子又はァセチル基であり、 X7—は対ィォ ンであり、 脱ァセチル化度は 50 100%、 好ましくは 70 L 00% q26は 20 3000、 好ましくは 50
1000である。 なお、 式中の一 N + H2R66基の N 原子とァ 二オンボリマ一のァニオン基とが結合する。
( 2 ) 中性多糖類のジェチルアミノエチル誘導体:
中性多糖類としては、 デキストラン、 セルロース、 マンナン、 スターチ又はァガロース等を挙げることができる。 これら誘導 体のジェチルアミノエチル置換度は、 糖残基 1個当り 0 . 5〜 2 . 0基、 好ましくは 0 . 7〜1 . 5基であり、 重合度は、 5 0〜 5 0 0 0、 好ましくは 1 0 0〜; L 0 0 0である。 なお、 ジェチルァミノエチル基の窒素原子とァニオンボリマーのァニ オン基とが結合する。
前記一般式(IV ) のァニオンポリマー (b 1 ) の具体例を拳 げれば、 ポリグルタミン酸、 ポリアスパラギン酸又はこれらポ リマーを構成する単量体のコポリマー、 更に、 これら単量体と グリシン、 ァラニン、 フエ二ルァラニン、 チロシン、 バリン、 ロイシン、 イソロイシン、 セリン、 トレオニン、 メチォニン、 システィン、 ヒスチジン、 プロリン、 及び/又はトリプトファ ンなどとのコポリマーである。
なお、一般式(III)及び(IV)で示されるポリアミノ酸は、 一般的な酸無水物モノマー法、 活性エステル化法、 メリーフィ ールド法等によって合成することができる。
前記一般式(V )のァニオンポリマー (b 2 ) の具体例を挙 げれば、 ポリアクリル酸、 ボリメタクリル酸、 ポリイタコン酸 モノエステル、 ポリマレイン酸モノエステル、 ポリビニルスル ホン酸、 ポリスチレンスルホン酸、 これらポリマーを構成する 単量体のいずれか 2種以上のコボリマー、 更に、 これら単量体 とその単量体のカルボキシル基にエステル結合によって結合し た炭素数 6〜 1 8個のアルキル基を有するカルボン酸誘導体と のコポリマーである。

本発明で用いる高分子電解質錯体( P E C ) は通常の方法で 調製することができる。 即ち、 前記のカチオンボリマー及びァ 二オンポリマーの各水溶液( 10—5モル リットル〜 10—2モ ル Zリットル) を、 カチオンポリマーのカチオン席とァニオン ポリマーのァニオン席との濃度比(カチオン席ノア二オン席) が 0. 25〜4. 0の範囲内、 好ましくは 0. 4〜2. 5の範 囲内で、 水溶液中で反応させる。 カチオン席とァニオン席との 濃度比(カチオン席 Zァニオン席) が 0. 25〜4. 0の範囲 外になると、 高分子電解質錯体(PEC)が形成され難くなる ので好ましくない。 この反応は比較的活性が高いので、 溶液の pH、 イオン強度、 温度などは比較的広い範囲であることができ るが、 一般的には pH3〜9、 イオン強度 0〜1. 0及び 20〜 4 CTCで実施する。
本発明で用いる P E Cの荷電バランスは一 6〜十 6、 好まし くは一 4. 5〜+ 4. 5である。 ここで荷電バランスとは、 P E Cの荷電状態を、 その出発材料であるカチオンボリマー及び ァニオンポリマーの、 各々のカチオン席及びァニオン席の濃度 比で表現するものである。 例えば、 使用するカチオンポリマー のカチオン席及びァニオンポリマーのァニオン席の鴿度が等し い場合は、 生成する PECの荷電バランスは ±0となる。 濃度 比がこれより大きければ(即ち、 カチオン席の漉度の方が高け れば)荷電バランスは正となり、 小さければ(即ち、 ァニオン 席の濃度の方が高ければ) 負となる。 また、 濃度比が 1 . 5の 場合は荷電バランスは + 2となり、 濃度比が 0 . 5の場合は荷 電バランスは一 3 . 3となる。 荷電バランスの調整は、 等濃度 のカチオンポリマー水溶液及びァニオンポリマー水溶液の混合 量を変化させることによつて容易に行なうことができる。 荷電 バランスの調整により、 カチオン過剰又はァニオン過剰の状態 とすることができる。
高分子電解質錯体(P E C ) は、 反応液からゲル状の沈殿物 として得られるので、 そのゲル状沈殿物のままで、 又はそのゲ ル状沈殿物から直接に成形 ·加工して適当な形状(例えば、 繊 維状、 フィルム状、 シート状、 塊状、ラテックス状又はゲル状) とし、 そのまま湿潤状態又は乾燥状態の抗菌性材料として用い ることができる。 また、 P E Cはほとんどすベての材料に付着 又は接着させることができるので、 適当な担体にコーティング して、 抗菌性材料を調製することができる。 また、 用途によつ ては (液自体に抗菌性をもたせる場合) 、 P E C反応液をその まま (懸濁液) の状態で用いることもできる。
有機材料からなる担体としては、 例えば、 有機高分子材料、 例えば、 合成若しくは天然樹脂、 合成若しくは天然ゴム、 合成 若しくは天然繊維、 生体高分子材料、 皮革、 木材、 パルプ、 紙 を挙げることができる。
合成樹脂としては、 例えば、 炭化水素重合体(例えば、 ポリ ォレフィン、 ポリエチレン、 ポリプロピレン、 ボリブテン一 1、 ポリ一 4ーメチルペンテン一 1、 スチレン樹脂、 ボリァセチレ ン) ;ハロゲン化炭化水素重合体(例えば、 ボリ塩化ビニル、 ポリ塩化ビニリデン、 フッ素樹脂〉 ;不飽和アルコール又はェ 一テル重合体 (例えば、 ポリビニルアルコール、 ボリビニルェ 一テル、 ポリフエ二レンォキサイ ド、 ポリフエ二レンサルファ イド、 ァセタール樹脂、 ポリエーテル、 ポリビニルプチラール、 ポリエーテルスルホンエポキシ樹脂) ;不飽和アルデヒド又は ケトン重合体 (例えば、 フエノール樹脂、 ユリア樹脂) ;不飽 和カルボン酸重合体 (例えば、 ポリアクリレート、 アクリル樹 脂) ;不飽和エステル重合体 (例えば、 ポリビニルエステル、 ボリァリレート、 全芳香族ポリエステル、 ポリエチレンテレフ タレート、 ポリカーボネート、 ボリプチレンジァリルフタレー ト、 不飽和ボリエステル樹脂) ;不飽和二トリル重合体 (例え ば、 ポリアクリロニトリル、 A B S樹脂、 A A S樹脂、 A E S 樹脂) ;不飽和ァミン重合体 (例えば、 ポリビニルァミン、 ポ リイミド、 ポリアミド、 メラミン樹脂、 ポリエチレンィミン、 ボリウレタン〉 ; その他、 シリコーン樹脂、 上記の共重合体、 ブレンド樹脂、 熱可塑性ェラストマーなどを挙げることができ る。 天然樹脂としては、 例えば、 繊維素誘導体樹脂を挙げるこ とができる。
合成ゴムとしては、 例えば、 スチレン一ブタジエンゴム、 ブ タジェンゴム、 イソプレンゴム、 二トリ口ゴム、 クロ口プレン ゴム、 ブチルゴム、 エチレン一プロピレンゴム、 ァクリルゴム、 塩素系ボリエチレンゴム、 フッ素ゴム、 シリコーンゴム、 ウレ タンゴム、 多硫化ゴムを挙げることができる。
合成繊維としては、 例えば、 再生セルロース (ビスコースレ 一ヨン、 キュフ。ラ) アセテート、 トリアセテート、 ポリアミド、 アクリル、 ビニロン、 ビニリデン、 ボリ塩化ビニル、 ポリエス テル、 ポリエチレン、 ポリプロピレン、 ポリべンゾエート、 ポ リクラーノレ、 ァラミドフエノール系繊維、 ポリウレタン系繊維、 フッ素繊維、 ポリビニルアルコール、 炭素繊維、 炭化硅素繊維 を挙げることができる。 天然繊維としては、例えば、 木綿、 絹、 羊毛、 麻、 木材を挙げることができる。
担体としては無機材料、 例えば、 ガラス、 鉱物(例えば、 石 線) 、琺瑯、セメント、 セラミックス、 人造石、 金属 (例えば、 鉄、 鋼、 非鉄金属、 合金) を用いることもできる。 担体の形状 及び形態は特に制限されず、 繊維状、 フィラメント状、 フィル ム状、 シート状、 織布、 不織布、 棒状、 紐状、 球状、 粉体、 粒 体、 多孔質体、 中空体、 凝集体、 発泡体、 ゲル状体等、 いかな るものでもよい。
P E Cを担体に担持させるには、 任意の公知の方法、 例えば、 塗布、 噴霧又は浸漬などの方法で行なうことができる。 P E C 溶液を担 こ単に接触させるだけでもよい。 例えば、 カチオン ポリマー水溶液とァニオンポリマー水溶液とを反応容器内で混 合した後、 反応溶液を直ちに所定の容器に移し、 一昼夜程度静 置して P E Cを十分に析出させた後、 上澄液を除去し、 生理食 塩水及び蒸留水で 1〜 3回程度洗浄し、 6 0〜 1 0 0 °Cで 6〜 1 2時間乾燥してアニーリングすると、 その容器の底面に P E Cを強固に付着させることができる。 その容器の内面全体をコ 一ティングするには、 一昼夜程度回転振盪して P E Cを十分に 析出させた後、 上澄液を除丟し、 前記と同様に洗浄し、 ァニー リングすると P E Cを容器面に強固に付着させることができる。 また、 繊維状、 ビーズ状又は織布状等の材料の場合には、 そ れらの材料を P E C液に一昼夜程度浸漬し、 続いて前記と同様 に処理すればよい。 また、 特開昭 5 0— 6 3 0 9 6号公報に記 載されているように、 P E C合成を水可溶性有機溶媒 (例えば、 水とアセトンと臭化ナトリウムとの混合物) の存在下に行ない、 反応液をそのまま塗料として用いて塗布、 噴霧、 又は浸漬する こともできる。
P E Cを担持させる担体表面の疎水性が高い場合 (例えば、 ポリカーボネート製担体〉 には、 その表面に親水性を付与する 処理 (次亜塩素酸処理、 有機溶媒処理、 プラズマ処理、 紫外線 処理など) を予め行なうのが好ましい。
こうして得られた P E C担持担体は抗菌性材料としてそのま ま利用するか、 あるいはその抗菌性材料を用いて各種の抗菌性 製品を調製することができる。 そのまま利用することのできる 抗菌性材料としては、 繊維材料 (例えば、 繊維、 フィラメント、 織布、 不織布) の表面の少なくとも 1部分 (好ましくは全体) に P E Cを担持させたもの、 例えば、 P E Cを担持したガーゼ、 脱脂綿、 生地 (医療、 衛生若しくは美容用の無菌衣料製品用) を挙げることができる。 これらの P E C担持繊維材料から、 例 えば、 マスク、 眼帯、 包帯、 シーツ、 吸収パッド (例えば、 耳 用、 鼻用、 口腔用若しくは月経用タンポン) 、 ナプキンを簡単 に調製することができる。
更に、 各種の無菌衣類、 例えば、 下着 (シャツ、 肌着、 靴下 類など) 、 ベビー用リンネル製品 (例えば、 ベビー用パンツ、 よだれ掛け、 産着、 胴着など) 、 ハンカチ、 コルセット、 ガー ドル、 ブラジャー、 海水着、 手術用衣服、 外科医用又は患者用 エプロン、 救命具、 潜水服、 実験室着、 保護着 (手術用手袋) 、 マスク、 手術用帽子を調製することもできる。 P E C担持抗菌性材料を用レ、て調製する抗菌性製品としては、 微生物の増殖を抑制することが好ましい製品であれば特に制限 されるものではない。 例えば、 医療関連器具類、 衛生鬨連器具 類(例えば、 病院用べッドカバー、 シーツ、 無菌衣服、 包帯、 おむつ、 眼帯ガーゼ、 タンポン、 コンタクトレンズ、 コンタク 卜レンズ容器、 医薬品保存容器、 輸血用容器) 、 食品関連器具 (例えば、 食品用包装材料、 食品保存容器) 、 生话関連器具類 (例えば、 食卓用具、 台所用具例えば、 食器棚下敷きシート、 サニタリー用品例えば、便座カバー) 、 理美容器具類、 スライ ムが発生し易い器具(例えば、 透析膜、 沪過材) 、 その他理化 学機械器具(例えば加湿器、 洗浄器、 恒温槽)等を挙げること ができる。
本発明の P E C担持抗菌性材料を用いて調製する抗菌性製品 として特に好ましい医療鬨連器具類(好ましくは、 使い捨て用 医療鬨連器具類) について以下に列記する。 また、 それらの好 ましい担体材料を括弧内に示す。

カチオンポリマーのようなカチオン性高分子電解質では、 担 持されている対ィォンが一般に低分子量対ィォン(例えばハ口 ゲンイオン) であるので、 その対イオンがポリマーから脱離し てポリマー中のカチオン部位が露出することは比較的容易であ る。 これに対して、 本発明で用いる P E Cでは、 対イオンが相 互に高分子化合物であるので、 出発力チォンポリマーが有して いた第 4級アンモニゥ厶の性質は或る程度中和された形で存在 している。 こうした構造を有する P E Cが抗菌活性を示すこと は綮くべきことであり、 その理由は現在のところ解明されてい ないが、 ポリマー本体内に化学的に強固に結合して含有されて いる第 4級アンモニゥム部分が持続的な抗菌活性を示すものと 思われる。 更に、 一般に P E Cは、 親水性のミクロドメイン構 造、 表面の水の構造変化、 荷電バランスなどの変化によって著 しく多様な性質を示すので、 本発明においてもこれらの影響も 相俟って抗菌性を発現しているものと考えられる。 実施例
以下、 実施例によって本発明を更に具体的に説明するが、 こ れらは本発明の範囲を限定するものではない。 なお、 以下の実 施例に記載の平均分子量は蒸気圧降下法で測定した数平均分子 量である。
調製例 1 : PEC ( 2X-CLA) の調製
カチオンボリマーであるポリ [ (ジメチルイミニォ)ェチレ ン (ジメチルイミニォ) メチレン一 1 , 4一フエ二レンヌチレ ンジクロライド] ( 2X) (平均分子量約 6000〉
0. 015 g (カチオン席として 1 X 10一4モル) とァニオン ポリマーであるァクリル酸 Zラウリルァクリレートのランダム 共重合体(CLA) (アクリル酸含量約 80モル%;平均分子 量約 10, 000 ) 0. 018g (ァニオン席として 1 X
10— 4モル) とを、 それぞれ別々に生理食塩水 (pH7. 4 ) 1 Omlに溶解させ、 こうして得られた 2つの水溶液 5mlづっを ビーカー中にて一挙に混合させ、 高分子電解質錯体ゲル(荷電 バランス : ±0 ) を形成した。
前記と同様に調製したカチオンボリマー水溶液 7mlとァニォ ンボリマー水溶液 3 mlとをビーカー中にて一挙に混合させ、 高 分子電解質錯体ゲル(荷電バランス : +4 ) を形成した。
更に、 前記と同様に調製したカチオンポリマー水溶液 3m 1 とァニオンボリマー水溶液 7 mlとをビーカー中にて一挙に混合 させ、 高分子電解質錯体ゲル(荷電バランス: ー 4 ) を形成し た。
調製例 2 : PEC ( 2X-COA) の調製
カチオンボリマーであるポリ [ (ジメチルイミニォ)ェチレ ン (ジメチノレイミニォ) メチレン一 1 , 4—フエ二レンメチレ ンジクロライド] (2X〉 (平均分子量約 3000)
0. 015 g (カチオン席として 1 X 10— 4モノレ) とァニオン ポリマーであるァクリル酸 2—ェチルへキシルァクリレート のランダム共重合体(COA) (アクリル酸含量約 60モル%; 平均分子量約 8000 ) 0. 021 (ァニオン席として 1 X10一4モル) とを、 それぞれ別々に蒸留水(pH8. 0 ) 1 Omlに溶解させ、 こうして得られた 2つの水溶液 5mlづっを ビーカー中にて一挙に混合させ、 高分子電解質錯体ゲル(荷電 バランス: ±0 ) を形成した。
前記と同様に調製したカチォンポリマー水溶液 7 mlとァニォ ンポリマ一水溶液 3 mlとをビーカー中にて一挙に混合させ、 高 分子電解質錯体ゲル(荷電バランス: +4) を形成した。

調製例 9 : PEC 「ポリ (L一リジン) — CLA] の調製
カチオンポリマーであるポリ (L一リジン) (PLL) (平 均分子量約 3000) 1. 3mg (カチオン席として 1 X 10_5 モル) とァニオンポリマーであるァクリル酸 Zラウリルァクリ レートのランダム共重合体(CLA) (アクリル酸含量約 80 モル%;平均分子量約 5000 ) 1. 8mg (ァニオン席として 1X10— 5モル) とを、 それぞれ別々に 0. 5モル /リットル の塩化ナトリゥム水溶液(pH6. 5 ) 10mlに溶解させ、 こう して得られた 2つの水溶液 5 mlづっを室温でビーカー中にて一 挙に混合させ、 高分子電解質錯体ゲル〔PLL— CLA〕 (荷 電バランス: ±0 ) を形成した。
前記と同様に調 1¾したカチオンポリマー水溶液 7 mlとァニォ ンポリマー水溶液 3mlとをビーカー中にて一挙に混合させ、 高 分子電解質錯体ゲル(荷電バランス: +4) を形成した。

カチオンポリマーであるリジン Zセリンのランダム共重合体 CC (Ly s/Se r) (リジン含量約 70モル%;平均分 子量約 10000 ) 0. 01 g (カチオン席として 1 X

調製例 12 : PEC (リジン セリン共重合体一カルボキシメ チルキチン) の調製

前記と同様に調製したカチオンポリマー水溶液 7mlとァニォ ンポリマー水溶液 3 mlとをビーカーにて一挙に混合させ、 高分 子電解質錯体ゲル(荷電バランス: +4 ) を形成した。
調製例 1 7 : PEC (キトサン— CLA) の調製
カチオンポリマーであるキトサン(脱ァセチル化度 70%) (平均分子量約 3000 ) 0. 02 1 g (カチオン席として 1 X 1 0一4モル) とァニオンボリマーであるァクリル酸 ゥラ リルァクリレートのランダム重合体 (C LA) (アクリル酸含 量約 80モル%) (平均分子量約 4000 ) 0. ◦ 18 g (ァ 二オン席として 1 X 10一4モル) とを、 それぞれ別々に 0. 3 モル/リットルの塩化ナトリウム水溶液(pH6. 5 ) 1 0mlに 溶解させ、 こうして得られた 2つの水溶液 5mlづっを室温でビ 一力一中にて一挙に混合させ、 高分子電解質錯体ゲル(キトサ ンー CLA) (荷電バランス : ±0 ) を形成した。
前記と同様に調製したカチオンポリマー水溶液 7 mlとァニォ ンボリマー水溶液 3mlとをビーカーにて一挙に混合させ、 高分 子電解質錯体ゲル(荷電バランス: +4 ) を形成した。
固定例 1 :担体(ポリエチレン製チューブ)への固定
調製例 1〜17で調製した P ECゲルそれぞれ 2mlを、 ポリ エチレン製チューブ(内径 1. 0cm〉 に分注し、 ローターで 60 r pmで 8時間回転させた。 ポリエチレン製チューブの内 壁に PECをコーティングした後、 上澄液を除去し、 ら (TCで 4時間乾燥し、 蒸留水 10mlで 3回チューブ内を洗浄し、 更に 8 CTCで 4時間乾燥させ、 PEC固定化チューブを得た。
固定例 2 :担体(ガーゼ)への固定
調製例 1〜17で調製したそれぞれの PECゲル液に、 木綿 ガーゼ( 1 OcmX 1 Ocm) を 8時間浸漬した。 ガーゼを取り 出して蒸留水 2 Omlで洗浄し、 6 TCで 4時間乾燥させ、 更に 蒸留水 1 Omlで 3回洗浄し、 8 OeCで 4時間乾燥させ、 PEC 固定化ガーゼを得た。
固定例 3 :担体(ガラスビーズ)への固定
調製例 1〜 6で調製したそれぞれの P E Cゲル液に、 ガラス ビーズ(直径 0. 2mm:東芝バロティー二社製) を 6時間浸 漬した。 ガラスビーズに PECをコーティングした後、 上澄液 を除去し、 6 CTCで 4時間乾燥し、 蒸留水 1 Omlで 3回ビーズ を洗浄し、 更に 8 CTCで 4時間乾燥させ、 PEC固定化ビーズ を得た。
固定例 4 :担体〔コンタクトレンズ材質(ポリメチルメタクリ レート) 〕への固定
調製例 7〜17で調製したそれぞれの PECゲル液に、 コン タクトレンズと同材質のボリメチルメタクリレート板を直径 8 mmに打ち抜いた円形小片(以下円形小片ともいう) を室温で 10時間浸漬した。 PECをコーティングした後、 上澄液を除 去し、 60でで 3時間乾燥し、 蒸留水 10mlで 2回、 生理食塩 水で 2回洗浄し、 更に、 8 (TCで 4時間乾燥させ、 PEC固定 化円形小片を得た。
蘂理試験例 1 :抗菌性の確認
以下の微生物を用いて抗菌性を調べた。
大腸菌( Escherichia coli) ATCC25932
ブドゥ球菌 ( Staphylococcus aureus ) ATCC25923
霊菌 ( Serratia marcescens ) IFO3046
緑腺 ¾ ( Pseudomonas aeruginosa ) fCC10145
ブレインハートインフュージョン ( BH I )培地で 37でに て 16時間培養した各種の菌液を、 0. 85%食塩含有 M/ 15リン酸緩衝液( P B S、 pH6. 8 )で希釈し、 菌濃度約 1 X 104假 Zmlの菌浮遊液を調製した。 PD培地(リン酸二力 リウム 7. 0 g、 リン酸一カリウム 2. 0 g、 硫酸マグネシゥ ム 0. 1 g、 硫酸アンモニゥム 1. 0 g、 クェン酸ナトリウム 0. 5 g、 ブドウ糖 10. 0 g、 及びバクトペプトン 10 · 0 gを精製水 1000mlに溶解したもの) 10mlを前記固定例 1 で調製した P ECコーティングポリエチレン製チューブに添加 し、 前記の各菌浮遊液 0. 1mlを加えて混合した後、 3 CTCに 保温した。 各チューブを 3 CTCで 24時間振盪培養し、 その培 地の濁りを目視観察して抗菌効果を確認した。 結果を表 1に示 す。
蓥理試験例 2 :抗菌性の確認
PD培地 50mlを坂口フラスコに取り、 固定例 2で得た PE C固定化ガーゼ( 6cmX 6cm) 3枚をそのフラスコに入れ、 前記薬理試験例 1と同様の各菌浮遊液 lmlを用いて約 1 X
104個植菌した。 同様 、 空試験用フラスコ及び PECを固 定していないガーゼ( 6cmX 6 cm) 3枚をフラスコに入れ、 前記と同様に植菌したものも用意した。 各フラスコを 3 CTCで 16時間振盪培養し、 その培地の濁りを目視観察して抗菌効果 を確認した。 結果を表 2に示す。
薬理試験例 3 :抗菌性の確認
PD培地 1 Omlを滅菌ポリエチレン製チューブに取り、 固定 例 3で得た PEC固定化ガラスビーズ 1 gを添加し、 前記薬理 試験例 1と同様の各菌浮遊液 0. lmlを加え混合した後 30°C に保温した。 同様に、 空試験用チューブ及び PECを固定して いないガラスビーズ 1 gを添加し、 前記と同様に植菌したもの も用意した。 各チューブを 30eCで 24時間振盪培養し、 その 培地の濁りを目視観察して抗菌効果を確認した。 結果を表 3に 示す。
薬理試験例 4 :抗菌性の確認
PD培地 1 Omlを滅菌ポリエチレン製チューブに取り、 固定 例 4で得た P E C固定化円形小片 2 gを添加し、 前記薬理試験 例 1と同様の各菌浮遊液 0. lmlを加えて混合した後、 3 CTC に保温した。 同様に、 空試験用チューブ及び P ECを固定して いない円形小片 2 gを添加し、 前記と同様に植菌したものも用 意した。 各チューブを 3 CTCで 24時間振盪培養し、 その培地 の濁りを目視観察して抗菌効果を確認した。結果を表 4に示す。 薬理試験例 5 :抗菌効果の持続性の確認
固定例 1で調製した各 P E C固定化チューブに精製水 1 Oml を加え、 振盪器で 2分間激振した後、 洗浄液を捨てた。 同様の 洗浄操作を更に 4回(合計 5回)行ない、 各チューブを 6 0 eC で 2時間乾燥した。 こう,して前処理したチューブを用い、 大腸 菌(ATCC25932 ) により薬理試験例 1と同様の操作で抗菌効 果の持続性を調べた。 結果を表 5に示す。
薬理試験例 6 :抗菌効果の持続性の確認
固定例 2で調製した各 P E C固定化ガーゼを 1 0 0 0 mlのビ 一力一に入れ、 精製水 5 0 0 mlを加え、 マグネットスターラー を用いて 5分間撹拌した後、 洗浄液を捨てた。 同様の洗浄操作 を更に 4回 (合計 5回)行ない、 各ガーゼを取り出し、 6 CTC で 4時間乾燥した。 こうして前処理したガーゼを用い、 大腸菌 (ATCC25932 ) により薬理試験例 2と同様の操作で抗菌効果 の持続性を調^^た。 結果を表 6に示す。

固定例 3で調製した各 P E C固定化ガラスビーズを滅菌ポリ エチレン製チューブに入れ、 精製水 1 0 mlを加え、振盪器で 2 分間激振した後、 洗浄液を捨てた。 同様の洗浄操作を更に 4回 (合計 5回)行ない、 各ガーゼを取り出し、 6 CTCで 4時間乾 燥した。 こうして前処理したチューブを用い、 大腸菌
(ATCC25932 ) により薬理試験例 3と同様の操作で抗菌効果の 持続性を調べた。 結果を表 7に示す。
薬理試験例 8 :抗菌効果の持続性の確認
固定例 4で調製した各 P E C固定化円形小片を滅菌ポリエチ レン製チューブに入れ、 精製水 1ひ mlを加え、 振盪器で 2分間 激振した後、 洗浄液を捨てた。 周様の洗浄操作を更に 4回 (合 計 5回)行ない、 円形小片を取り出し、 6 TCで 4時間乾燥し た。 こうして前処理した円形小片を用い、 大腸菌
(ATCC25932 ) により薬理試験例 4と同様の操作で抗菌効果 の持続性を調べた。 結果を表 8に示す。
蘂理試験例 9 :抗菌性の確認
調製例 1〜17で調製したそれぞれの P ECゲル液に、 沪紙 (T〇YO、 No. 5B) を 8時間浸漬した。 浐紙を取り出し て蒸留水 20mlで洗浄し、 6 CTCで 4時間乾燥させ、 更に蒸留 水 10mlで 3回洗浄し、 8 TCで 4時間乾燥させた。 得られた 沪紙を直径 13mmの円盤状に切り抜き、 ガス滅菌処理を行い、 後記の抗菌効果試験用ディスクとした。 一方、 P ECゲル液に 浸漬していない同様の円盤状沪紙を同様にガス滅菌処理して対 照用ディスクとした。
薬理試験例 1に記載の 4種の菌を、 薬理試験例 1と同様の方 法で B H I培地で一夜振盪培養し、 B H I培地で 3回遠心処理 した後、 同じく BH I培地で希釈し、 菌濃度約 1 X 107個ノ mlの菌浮遊液を調製した。 この菌浮遊液 20 1を前記の抗菌 効果試験用ディスク及び対照用ディスクに接種し、 37でで 2 時間放置した後、 トリプトソーャ寒天平板上に、 ディスク接種 面と寒天平板とを接するように置き、 37 Cで 1時間放置して から、 各ディスクを取り除いた。 更に、 37 Cで一昼夜培養を 行った後、 平板上に出現したコロニー形成を観察した。 結果を 表 9に示す。
実施例 1〜 1 1
前記の各調製例と同様にして各種の P ECを調製し、 担体に 固定してから薬理作用を調べた。 即ち、 表 10に示す各カチォ ンボリマーと表 1 1に示す各ァニオンボリマーとをそれぞれ表 10及び表 1 1の「採取量」欄に示す量で表 10の 「溶媒」橱 に示す溶媒 1 Omlに各々溶解させ、 得られた各溶液 5 mlづっを ビーカー中で一挙に混合させて高分子電解質錯体ゲル(荷電バ ランス: ±0 ) を形成した。 同様にカチオンボリマー溶液 7 ml とァニオンポリマー溶液 3 mlとから荷電バランス + 4、 そして カチオンポリマー溶液 3mlとァニオンポリマー溶液 7mlとから 荷電バランスー 4の各高分子電解質錯体ゲルを形成した。 これ らを固定例 2と同様の方法で担体(ガーゼ)へ固定し、 薬理試 験例 2と同様の方法で抗菌性を確認した。 それらの結果を表 12に示す。 なお、 表 10及び表 11では、 各ポリマーを略号 で示す。 それらの意味は以下の通りである (前記調製例で用い た略号は同じ意味であるので説明を除く) 。

1 . 繰返し単位中に 子を含有するカチオンポリマーと繰 返し単位中に一 C O 0—基、 一 S〇3一基又は一 P〇3—基を含有 するァニオンボリマーとを反応させることによって得られる高 分子電解質錯体を含有することを特徴とする抗菌剤。
2 . カチオンポリマーのカチオン席とァニオンポリマーのァニ オン席との濃度比 (カチオン席 Zァニオン席) が 0 . 2 5〜 4 . 0である請求項 1記載の抗菌剤。
3 . カチオンポリマーとして、
( a 1 ) 一般式 ( I )
[式中、 R i及び R 4はそれぞれ独立に炭素数 l 1 0個のァ ルキレン基、 一般式
一 R
R 1 2 - (式中、 Rll及び Rl2は、 それぞれ独立に、 炭素数 1又は 2 個のアルキレン基である)
で表される基又はァリーレン基であって、 R2、 R3、 R5及び
Γ 6はそれぞれ独立に炭素数 1〜 3個のアルキル基であるか、 又は Riは式中の 2個の窒素原子及び R2、 R3、 R5及び R6 と一緖になって式
で表される基であって R4は前記と同じ意味であるものとし そして XI—は対イオンであり、 mは 5以上の数である] で表される化合物、

(式中、 vは 3又は 4であり、 R91は水素原子;炭素数 1〜 4個のアルキル基; ヒドロキシ基、 メルカプト基若しくは炭素 数 1〜3個のアルキルチオ基で置換された炭素数 1〜4個のァ ルキル基;又はイミダゾリルメチル基若しくはインドリルメチ ル基であり、 R92は— N + H2X5—又は一 N+HC (NH) N H 2 X 6—であり、 X 5 _及び X 6—はそれぞれ独立に対ィォンで あり、 tは 20〜10◦であり、 そして rは 10以上の整数で ある)
で表される化合物、 及び
( a4 ) カチオン性多糖類
からなる群から選んだ少なくとも 1種の化合物を用いる請求項 1記載の抗菌剤。
4. ァニオンポリマーとして、
(b l )—般式(IV)
(式中、 uは 1又は 2であり、 R81は水素原子;炭素数 1 4個のアルキル基; ヒドロキシ基、 メルカプト基若しくは炭素 数 1〜 3個のアルキルチオ基で置換された炭素数 1〜4個のァ ルキル基; 又はィ ミダゾリルメチル基若しくはインドリルメチ ル基であり、 sは 20〜100であり、 そして qは 10以上の 整数である)
で表される化合物、
(b 2〉 一般式 (V)
(式中、 R31は水素原子又はメチル基であり、 R 32は炭素数
6〜18個のアルキル基であり、 Yはカルボン酸基若しくはそ の塩、 スルホン酸基若しくはその塩、 又はスルホン酸基若しく はその塩を含有するァリール基であり、 aは 20〜100の数 であり、 Pは 1 0以上の数である)
で表される化合物、 及び
(b 3 ) ァニオン性多糖類
からなる群から選んだ少なくとも 1種の化合物を用いる請求項 1記載の抗菌剤。
5. 前記一般式 ( I ) において Ri及び R4がそれぞれ独立に 炭素数 2〜 8個の直鎖又は分枝鎖のアルキレン基であり、 Rl 1及び Ri 2が P位で結合している前記一般式 ( I ) で表さ れる化合物 ( a 1 〉 を用いる請求項 3記載の抗菌剤。
6. 前記一般式(III) において R91が水素原子、 メチル基、 イソプロピノレ基、 イソブチル基、 s—ブチル基、 ヒドロキシメ チル基、 ヒドロキシェチル基、 メチルチオェチル基、 メルカプ トメチル基、 5—イミダゾリルメチル基又は 3—イミダゾリル メチル基である前記一般式(III) で表される化合物 ( a 3 ) を 用いる請求項 3記載の抗菌剤。
7. カチオン性多糖類(a4) としてキトサン、 キトサンの誘 導体、 又は中性多糖類のジェチルアミノエチル誘導体を用いる 請求項 3記載の抗菌剤。
8. 前記一般式(IV) において R81が水素原子、 メチル基、 イソプロピル基、 イソブチル基、 s—ブチル基、 ヒドロキシメ チル基、 ヒドロキシェチル基、 メチルチオェチル基、 メルカプ トメチル基、 5—イミダゾリルメチル基又は 3—イミダゾリル メチル基である前記一般式(IV) で表される化合物(b 1 ) を 用いる請求項 4記載の抗菌剤。
9. 前記一般式(V) において R32が炭素数 6〜14個の直 鎖若しくは分枝鎖のアルキル基であり、 aが 50〜 100であ る前記一般式(V) で表される化合物(b2) を用いる請求項 4記載の抗菌剤。
10. ァニオン性多糖類としてヒアルロン酸及びその誘導体、 アルギン酸及びその誘導体、 コンドロイチン硫酸 A、 コンドロ ィチン硫酸 C、 コンドロイチン硫酸 B (デルマタン硫酸)及び その誘導体、 コンドロイチン硫酸 D及びその誘導体、 コンドロ ィチン硫酸 E及びその誘導体、 へパラン硫酸及びその誘導体、 へパリン及びその誘導体、 AT—力ラゲナン及びその誘導体、 λ 一力ラゲナン及びその誘導体、 カルボキシメチル化、 硫酸化又 はリン酸化したセルロース誘導体、 カルボキシメチル化、 硫酸 化又はリン酸化したキチン誘導体、 カルボキシメチルスターチ 及びその誘導体、 カルボキシメチル化、 硫酸化又はリン酸化し たアミロース誘導体、 カルボキシメチル化、 硫酸化又はリン酸 化したアミ口べクチン誘導体、 カルボキシメチル化、 硫酸化又 はリン酸化した /3— 1, 3 ' ーグルカン誘導体、 カルボキシメ チル化、 硫酸化又はリン酸化した 3— 1, 2 ' —グルカン誘導 体、 カルボキシメチル化、 硫酸化又はリン酸化した/ S— 1 , 3 ' - ; /3 - 1 , 6 ' ーグルカン誘導体、 カルボキシメチル化、 硫 酸化又はリン酸化したデキストラン誘導体、 カルボキシメチル ィ匕、 硫酸化又はリン酸化したプルラン誘導体、 カルボキシメチ ル化、 硫酸化又はリン酸化したァガロース誘導体、 カルボキシ メチル化、 硫酸化又はリン酸化した /3— 1 , 4 ' 一ガラクタン 誘導体、 カルボキシメチル化、 硫酸化又はリン酸化したマンナ ン誘導体、 及びカルボキシメチル化、 硫酸化又はリン酸化した ィヌリン誘導体からなる群から選んだ少なくとも 1種のァニォ ン性多糖類 ( b 3 ) を用いる請求項 4記載の抗菌剤。
1 1 . 請求項 1〜1 0のいずれか一項に記載の高分子電解質錯 体を担体上に担持することを特徴とする抗菌材料。
