Abstract
Verfahren
zur Identifizierung und Quantifizierung von Mikroorganismen in einer
Probe oder einer Lebensmittelprobe, aufweisend folgende Verfahrensschritte:
– Isolierung
der Mikroorganismen aus der Probe,
– Überführung der isolierten Mikroorganismen
in ein PCR-Gefäß,
– Amplifikation
der Zielnukleinsäure
durch eine real-time PCR als Marker für die Mikroorganismen,
– Nachweis
der amplifizierten Zielnukleinsäure
mittels einer Sonde mit mindestens zwei Fluorophoren, die ein FRET-Paar
bilden, wobei die Fluorophoren so gewählt sind, daß erst nach
der Hybridisierung der Sonde mit dem Zielnukleotid ein erkennbares
Signal von dem Fluorophor emittiert wird, dass eine gegenüber der
Fluoreszenzemission der Probenumgebung verlängerte Lebensdauer aufweist,
so dass das Meßsignal
durch eine zeitaufgelöste Fluoreszenzmessung
von der Autofluoreszenz der Probenumgebung differenzierbar ist,
– Erfassen
der Fluoreszenzemission mittels zeitaufgelöster Fluoreszenzmessung,
– Quantifizierung
der Pathogen-Konzentration in der Ausgangsprobe über eine Standardkurve mit
